How does reference-standard error propagate into a dulaglutide content figure?

I am comparing a supplier certificate against an independent result on the same lot.

I have worked this out and I would like someone to find the error, because I suspect there is one.

My working so far, for the record, is below, and I am fairly sure the error is in the unit conversion rather than the algebra.

Is my approach right even if my number is wrong?

The relevant detail is that the cost of a certified standard is cheaper than the cost of one wrong conclusion based on an unverified standard, which is the argument for using them.

The response factor changes if the method changes — a different gradient time, a different temperature, a different column — so the standard must be run under the exact method conditions as the sample.

It helps to be literal here: storing a standard in a freezer is better than storing it at room temperature, but how much better depends on the chemical stability of the standard.

The pharmacopoeial standards specify system suitability requirements for quantitative methods that include replicate injection agreement and that agreement should be tighter than two per cent.

The limitation is that a standard only establishes a baseline; it does not establish that your method is correct.

Do not use an in-house standard without independent verification of its purity.

Mechanically, a standard should be stored as carefully as the samples you test it against, because a degraded standard skews all of your results in a consistent direction.

The single highest-value check is verifying that the standard's certificate specifies its purity and gives a traceable source.

To be exact about it, system suitability for a standard includes that replicate measurements of the same standard give consistent results, and if they do not, something in the method or the instrument is wrong.

Certified reference materials are available from national metrology institutes and carry full documentation of their purity and traceability.

The caveat is that a standard certificate is only as good as the issuing laboratory is reliable, and not all certificates are equally credible.

The practical summary: use a standard of declared purity and verified traceability, and check that the certificate is current.

The relevant detail is that using different standards for different samples is a recipe for systematic disagreement, which is why validation requires that the same standard be used for the entire series.

Using a standard that is older than its certificate validity window is using a material of unknown purity, which defeats the purpose of having a standard.

Concretely, comparing your quantitation result to a standard certificate requires that the sample purity be accounted for — if you measure a 95 per cent pure sample against a 99 per cent pure standard, the content result is biased.

Ask for the standard's purity when you buy it.

Mechanically, using a standard of unknown or undeclared purity is like building on sand — every number downstream inherits that uncertainty and amplifies it.

Purity of the standard is not the same as traceability of the standard — purity tells you the composition, traceability tells you the standard came from a credible source.

Certified reference materials are available from national metrology institutes and carry full documentation of their purity and traceability.

Do not use an in-house standard without independent verification of its purity.

Start from the certificate on the standard itself, because that tells you the uncertainty in every result that uses that standard.

Most research quantitation work assumes the sample and standard are treated identically through every step, which is almost never true if they come from different sources.

The pharmacopoeial standards specify system suitability requirements for quantitative methods that include replicate injection agreement and that agreement should be tighter than two per cent.

I would not accept a standard that does not come with a certificate stating its purity.

The practical summary: use a standard of declared purity and verified traceability, and check that the certificate is current.
